Optimization of enzymatic hydrolysis of antlers polypeptide by central composite design-response surface methodology

Abstract:

The optimization of enzymatic hydrolysis of antlers polypeptide was conducted by central composite design-response surface methodology. The antlers polypeptide was obtained by pepsin hydrolysis antlers collagen which was decocted extract from antlers. The effects of substrate concentration, enzyme dosage, temperature, pH and enzymolysis time on the peak areas and polypeptide content of peptides were investigated by single factor method. Then, substrate concentration, enzyme dosage and temperature were taken as the independent variables. Meanwhile, the polypeptide content of peptides and polypeptide peak areas were taken as the dependent variables for the central composite design-response surface test which could determine the best enzymatic hydrolysis process. Finally, the experiment was verified. The results indicated that the best processing conditions were: enzyme loading was 1.8%, substrate concentration was 0.08 mg/mL and the temperature was 40 ℃. The method was simple, feasible, stable and predictive.
