Extraction and enrichment of total alkaloids of Plumula Nelumbinis by ultrahigh pressureassisted micellar extraction combined with cloudpoint extraction

Abstract:

       We determined the technical conditions of ultrahigh pressureassisted micellar extraction by such singleelement experiment as surface active agent type, concentration, extraction pressure, time and the ratio of solid and liquid to construct the extraction and enrichment process of total alkaloids of Plumula Nelumbinis. We implemented the enrichment of the alkaloids of Plumula nelumbinis by NaCl addition, cloud point extraction and double water phase separation. The optimal extraction conditions are 5%(w/w) OP10 solvent, extraction pressure of 100 MPa, extraction time of 1 min and solid/liquid ratio of 1:20 (g/mL). The optimal extraction yield of total alkaloids is 103.6 mg/g. Its recovery rate is 70.73% and the preconcentration factor (CF) is 8.6 under the conditions of 15% NaCl addition, stability of 10 min at 50 ℃ and twophase separation. Results show that the extraction process is rapid and benefit for environment.
