Preparation of reference substances of vitexin, hyperoside and isoquercitrin from Crataegi Folium extract

Abstract:

Three low content flavonoids, such as vitexin, hyperoside and isoquercitrin from Crataegi Folium were rapidly separated and purified by highspeed countercurrent chromatography (HSCCC) combined with preHPLC. The total flavonoids from Crataegi Folium were firstly separated by HSCCC, with a solvent system of chloroform/methanol/water/ nbutanol (4∶3∶2∶1.5, V/V), flowrate of 5.0 mL/min, rotation speed of 850 r/min, detection wavelength of 254 nm. A mixed component was separated from crude flavonoids, containing vitexin, hyperoside, and isoquercitrin. Then, the mixture was separated by preHPLC and vitexin, hyperoside, and isoquercitrin were obtained, with the purity over 98%, respectively, as determined by HPLC. Experimental results show that it is an efficient method for separation of low content flavonoids with high purities from Crataegi Folium by HSCCC combined with pre-HPLC.
