Abstract: Lonicerae Japonicae Flos (LJF) is a traditional Chinese medicine known for its heat-clearing and detoxifying properties, with Shandong Province being a major production area. However, studies on differences in chemical composition among different regions within Shandong (especially saline-alkali lands) are limited. Furthermore, a single analytical technique is insufficient to comprehensively cover volatile and nonvolatile key components, thereby limiting origin traceability and quality evaluation. In this study, 13 batches of genetically similar LJF samples from four regions in Shandong—Guangrao (Dongying), Yuncheng (Heze), Yinan (Linyi), and Pingyi (Linyi)—were analyzed using a combination of three techniques, i.e., headspace gas chromatography–ion mobility spectrometry (HS-GC-IMS), headspace solid-phase microextraction–gas chromatography–mass spectrometry (HS-SPME-GC-MS), and ultra-performance liquid chromatography–quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS). Chemometric methods including one-way analysis of variance and orthogonal partial least squares–discriminant analysis (OPLS-DA) were employed to analyze compositional differences. A total of 267 compounds were identified, including five terpenoids unique to Guangrao, which are associated with saline-alkali stress. HS-GC-IMS detected 54 volatile compounds, with aldehydes, ketones, alcohols, and esters accounting for 87.0% of the total. HS-SPME-GC-MS analysis identified 16 differential components, nine of which were aldehydes. UPLC-Q-TOF-MS analysis identified 13 differential components. The OPLS-DA model demonstrated good stability (R²Y > 0.95, Q² > 0.89). This multitechnique approach provides a scientific basis for the quality evaluation and traceability of LJF in Shandong.
