复合酶膜生物传感器差分法检测牛乳中乳糖的含量

Determination of milk lactose content using composite enzyme membrane biosensor based on difference method

摘要: 采用生物传感器差分法,将半乳糖苷酶和葡萄糖氧化酶组成的复合酶以及葡萄糖氧化酶固定在过氧化氢双电极上,可同时测定样品中乳糖和葡萄糖的含量。结果表明,乳糖在0.00~1.50 g/L,葡萄糖在0.00~1.00 g/L内生物传感器法测定乳糖的线性良好,稳定性好,相对标准偏差为1.49%,加标回收率为95%~105%,本方法与高效液相色谱法比较,测定结果无明显差异(P>0.05)。因此,建立的复合膜生物传感器差分法可用于牛奶中乳糖的检测。

Abstract: The difference method associated with biosensor was used in this study by immobilizing glucose oxidase as well as composite enzyme consisting of galactosidase and glucose oxidase onto dual hydrogen peroxide electrodes. The levels of lactose and glucose in the sample could be simultaneously determined. The results showed that the biosensor method revealed a good linearity in the lactose range of 0.00~1.50 g/L and glucose range of 0.00~1.00 g/L,the stability of the biosensor was favourable, the relative standard deviation was 1.49%, and the adding standard recovery rate ranged from 95% to 105%,no significant difference (P>0.05) was noted in the measurement results between this method and HPLC. Therefore, the difference method associated with composite enzyme membrane biosensor proposed in this study can be used for the detection of lactose in milk.
