Research on negative impacts of vitrification of Salvia miltiorrhiza Bge.f.alba plantlets

Abstract:

          This paper addresses the impacts of different culture conditions on the vitrification of Salvia miltiorrhiza Bge.f.alba seedlings. We investigated the influences of KT/NAA ratio, active carbon and agar concentration on the prevention of vitrification of tissue culture seedlings with the tender stems of Salvia miltiorrhiza Bge. f.alba C.Y. Wu et H.W.Li.‘ buyu’ as the explants and MS as basic medium. Experimental results show that the tissue culture seedlings has higher bud multiplication coefficient and lower vitrification rate when KT concentration of 1.5 mg·L-1, NAA concentration of 0.5 mg·L-1, active carbon of 0.3% and agar concentration of 0.8% are employed. Therefore, the MS+KT 1.5 mg·L-1 +NAA 0.5 mg·L-1 + agar 0. 8% +active carbon 0.3% medium can effectively prevent vitrification of plantlets.
