丹参组培条件优化研究

Conditions optimization of tissue culture technique of Salvia Miltiorrhiza Bge.

摘要:

为优化丹参组培快繁体系,以带腋芽的丹参植株嫩茎为外植体,观察在不同组成培养基上和不同培养条件下的丛生芽诱导及生根情况,确定最佳培养条件。结果表明,MS+6BA(1.0 mg/L)+NAA(0.5 mg/L)+蔗糖(3×104 mg/L)+琼脂(7×103 mg/L)对丹参丛生芽诱导作用最好;1/2MS+IBA(1.0 mg/L)+KT(1.0 mg/L)+活性炭( 5×102 mg/L)+蔗糖(2×104 mg/L)+琼脂(7×103 mg/L)最有利于组培苗生根,组培苗的最佳生根温度为22 ℃。优化的丹参组培快繁体系,可用于丹参良种无性快繁,也可用于良种复壮及脱毒。

Abstract:

To optimize rapid propagation of tissue culture technique of Salvia miltiorrhiza Bge., we addressed the impact of its tissuecultured plantlets by different plant growth regulators and different culture conditions with tender stems with axillary bud as explants. Results show that the most effective induction to axillary buds is MS+6BA(1.0 mg/L)+NAA(0.5 mg/L)+sucrose(3×104 mg/L)+agar(7×103 mg/L). The most favorable rooting conditions of tissue culture seedling are 1/2MS+IBA(1.0 mg/L)+KT(1.0 mg/L)+activated carbon(5 ×102 mg/L)+sucrose(2×104 mg/L)+agar(7×103 mg/L) and 22 ℃ . This technique can be applied to rapid propagation, rejuvenation and detoxification of Salvia miltiorrhiza Bge.
