一株解蛋白芽孢杆菌的分离及生理特性研究

Isolation and physiological characterization of a bacillus proteolyticus strain

摘要: 从土壤中分离到一株可产胞外多糖(exopolysaccharides,EPS)的细菌,结合形态学特征和16S rDNA序列分析,鉴定为解蛋白芽孢杆菌,命名为LY0。实验发现,LY0分泌多种胞外酶、胞外多糖等胞外产物。抑菌圈实验表明,LY0可抑制包括大肠杆菌、金黄色葡萄球菌等多种细菌的生长。利用粘度计对液体培养的LY0菌株进行粘度测量,定时取样测定菌液的粘度,发现24 h时LY0菌液的粘度最高。随后,在发酵培养基中分别加入不同碳源培养24 h收集发酵液,结合透析袋和三氯乙酸纯化、利用苯酚–硫酸法测定多糖产量。实验结果显示,与其他碳源相比、以蔗糖为碳源产生的胞外多糖量最高,LY0产生了32.2 mg/L胞外多糖,说明蔗糖为LY0发酵生产胞外多糖的最适碳源。本实验所获得的解蛋白芽孢杆菌可用于微生物胞外多糖的生产,具有广泛的应用前景。

Abstract: Abacterial strain capable of producing exopolysaccharides (EPS) was isolated from soil and identified asBacillus proteolyticusstrainbased on morphological characteristics and 16S rDNA sequence analysis. The strain was designated as LY0. Experimental results showed that LY0 secreted various extracellular products, including extracellular enzymes and EPS. Antimicrobial activity, assessed by inhibition zone assays, showed that LY0 inhibited the growth of several bacteria such asEscherichia coliandStaphylococcus aureus. Viscosity measurements of LY0 liquid cultures revealed a peak at 24 h. Different carbon sources were added to the fermentation medium, and EPS yield was determined using the phenol–sulfuric acid method following purification with dialysis and trichloroacetic acid. The results showed that, compared with other carbon sources, sucrose yielded the highest EPS production (32.2 mg/L), suggesting that sucrose is the optimalcarbon source for EPS production by LY0. TheBacillus proteolyticusstrain obtained in this study can be used for microbial EPS production and holdsbroad application prospects.
