The influence factors of fluorescence determination of chlorophyll a and the data correction for them

摘要:

研究了浊度、温度对叶绿素a现场荧光仪测定叶绿素a的影响。分别用可见光分光光度计和叶绿素a现场荧光仪测定了不同温度、浊度梯度下小球藻水样的叶绿素a浓度值,探究影响规律和修正方法。结果表明,温度在7 ~ 30 ℃范围内,对叶绿素现场荧光仪测定叶绿素a浓度无显著影响;浊度在0 ~ 1 000 度范围内,需用叶绿素a浓度实测值减去0.0174倍的浊度值(度)对测定值进行修正。

Abstract:

The impacts of turbidity and temperature on the determination of chlorophyll a in the sea water with field fluorescence method were studied in this paper. The chlorophyll a concentration of Chlorella vulgaris at different temperature and turbidity gradients was determined by visible spectrophotometer and the field fluorescence respectively, to explore the influence rule and the method for correction. Results reveal that temperature has no significant effects on the concentration of chlorophyll a in the range of 7 ~ 30 ℃, while turbidity has significant influence on the determination of chlorophyll a in the range of 0 ~ 1 000 NTU, which should be modified by the following method: The true value equals to measured value minus 0.017 4 times’ turbidity value (NTU).
