牛膝中齐墩果酸和β-蜕皮甾酮的HPLC法含量测定及指纹图谱研究

Content determination of βecdysterone and oleanolic acid in Achyranthes bidentata Blume by HPLC and their fingerprints

摘要: 建立了中药牛膝的HPLC指纹图谱并同时测定其中齐墩果酸和β-蜕皮甾酮的含量。采用Agilent ZORBAX SBC18色谱柱,流动相为乙腈0.6%甲酸,柱温为25 ℃,检测波长为260 nm,进行了13批次牛膝药材的分析。牛膝药材的HPLC指纹图谱共有16个共有峰,4个特征峰,定量测定了其中两个峰β蜕皮甾酮和齐墩果酸。通过相似度分析初步断定,不同地域的牛膝药材在质量上存在明显的差异。该研究为建立牛膝药材的质量控制体系提供了方法和技术支持。

Abstract: We constructed quantitative high performance liquid chromatography (HPLC) fingerprint of Achyranthes bidentata Blume and determined the contents of oleanolic acid and βecdysterone. We analyzed 13 batches of Achyranthes bidentata Blume with Agilent ZORBAX SBC18 column, 0.6% formic acid as fluid phase, column temperature of 25 ℃ and detection wavelength of 260 nm. HPLC fingerprint of Achyranthes bidentata Blume included sixteen common peaks and four characteristic peaks. We also quantitatively determined two of these peaks, peak 5 and peak 7. From their similarity analysis, we primarily confirmed Achyranthes bidentata Blume from different origins had significant distinctions in quality. This research provides an approach and technology support for the quality control of Achyranthes bidentata.
