摘要: 本文研究了生物传感分析仪在共固定化GODCAT树脂的过氧化氢(H2O2)质量浓度的测定和酶活力的表征中的应用。首先,采用过氧化氢电极法检测了反应液中过氧化氢的质量浓度,过氧化氢由不同GOD/CAT酶活比条件下制备的共固定化GODCAT树脂催化氧化葡萄糖生成,从而找出了最适的GOD/CAT酶活比;然后,采用葡萄糖氧化酶电极法检测了催化氧化过程中反应液中底物葡萄糖质量浓度的变化,提供了一种共固定化GODCAT树脂的活力表征方法,并以此为基础研究了共固定化GODCAT树脂的酶学性质。结果表明,共固定化GODCAT树脂的最适GOD/CAT活力比是1:1;共固定化酶的最适温度是45 ℃,最适pH值为6.0,其热稳定性、pH稳定性、操作稳定性及贮存稳定性较游离酶均有一定的提高。

Abstract: We addressed the application of H2O2 concentration measurement and enzyme activity characterization in coimmobilized GODCAT resin. We initially detected H2O2 concentration in reaction solution with hydrogen peroxide electrode.H2O2was generated through coimmobilized GODCAT resin catalytic oxidizing glucose prepared in different GOD/CAT activity ratios. The optimal GOD/CAT activity ratio was then obtained. We further detected the change of glucose mass and concentration in the substrate of reaction liquid in catalytic oxidation with glucose oxidase electrode. This provided anactivity characterization method of coimmobilized GODCAT resin. We then investigated its enzymatic properties. Results show that its optimal generation conditions are GOD/CAT activity ratio of 1:1, temperature of 45 ℃ and pH of 6.0. Its heat, pH, operation and storage stabilitiesare all higher than those of free enzyme.
